Molecular analyses

Molecular analyses gather the following activities : NMR, Mass spectrometry, next generation sequencing and omics bioinformatics.

PIXANIM

The PIXANIM platform (Phenotyping by In / eX vivo Imaging from Animal to Molecule) is an experimental and analytical platform. PIXANIM enables animal phenotyping and functional exploration, from the whole animal to the molecule. PIXANIM is ISO 9001 certified, IBISA labeled and member of the Institut Carnot France Futur Elevage. Molecular analyzes with a set of technologies dedicated to proteomics and lipidomics for the identification and quantification of biomolecules.

Equipment : MALDI-TOF RapifleX mass spectrometer, LTQ Velos Pro Obitrap mass spectrometer. Three standard hospital operating theaters. Long-term sheep / pig accommodation rooms.

Mass spectrometry platform:

The platform specializes in the analysis of biomolecules (whole proteins, peptides, oligonucleotides, glycans, polymers or small organic molecules) which can be pure or in complex mixtures.

The Mass Spectrometry and Proteomics platform of the Center for Molecular Biophysics supports MALDI-TOF / TOF, nanoLC-IT MS / MS and ESI-UHR-QTOF (FR2708) analyzes. These analyzes can be carried out for different purposes:

- Check the quality of a product resulting from organic synthesis, biological engineering or biochemical purification (QC analyzes, characterization of biotherapeutic proteins and nucleic acids),

- Determine the crude formula of a compound,

- Identify unknown molecules (metabolism, impurity, etc.),

- Locate post-translational or induced modifications of proteins,

- Identify proteins (proteomics)

Equipment : 3 mass spectrometers. 1 MALDI-TOF / TOF UltrafleXtreme (Bruker), 1 ESI-Trappe Ionique HCT Ultra-ETD (Bruker), 1 UHR-ESI-QTOF MaXis-ETD (Bruker) and 1 UHR-Q-TOF MaXis-CID as part of the FR2708. ESI source spectrometers can be coupled to one of 3 liquid chromatographies: CapLC (Waters), UHPLC U3000 and nanoUHPLC U3000 (Dionex).

Analytical techniques platform:

The Analytical Techniques platform is mainly dedicated to the analysis and characterization of compounds resulting from organic synthesis.

Possible analyzes: reaction monitoring (FIA-MS, LC-MS & TLC-MS); measure of purity; quantification; spectrophotometric measurements.

Equipment : single quadrupole (MSQ, Thermo) + Ultimate 3000; triple quadrupole (Endura, Thermo) + Ultimate 3000RSLC; TLC-ESI-MS; HPLC-UV-DEDL (Agilent); GC-FID (Shimadzu)

Spectroscopic techniques : FT-IR ; UV-Vis ; Fluorescence ; RMN  400 Mhz, 250 Mhz.

BISCEm (Biologie Intégrative Santé Chimie Environnement)

Multidisciplinary technological platform of the GEIST research institute from Limoges University, bringing together the major families of technologies for the life sciences and allowing, within a single structure, the study of whole organisms, tissues, cells and molecules. BISCEm is organized into 3 activity poles which contribute to the advancement of the research programs of user teams with the following main research themes: infectious agents (bacteria, viruses, parasites), immunity, transplantation, cancer, neurosciences, and natural resources.

The molecular analyses division combines nuclear magnetic resonance (NMR - structural identification of small and medium natural or synthetic molecules in solution), mass spectrometry (identification and quantification of small molecules and proteins, omics applications), analysis nucleic acids (qualitative and quantitative analysis, high throughput real-time PCR, sequencing, omics applications), and bioinformatics which processes "omics" data. The pole works in a coordinated manner with the sequencing functional unit of the University Hospital Center and is developing a collaborative project with the PREMISS platform of the University of Limoges.

Equipment : BISCEm has more than 80 equipment for basic care and the performance of experiments in respect of animal welfare, and more than 30 semi-heavy and heavy equipment and dedicated software for cell, tissue and molecular analyses . The equipment is spread over nearly 1,300 m2 of dedicated premises on the Limoges university hospital campus. Other equipment is accessible via the 5 partner platforms and the site's research teams.

INRAE BioMAP research Team:

The "Anti-Parasitic BioMedicines" research team has a recombinant protein production platform, mainly focused on antibodies and derivatives (scFv, diabody, minibody, bispecific antibody, etc.).

The expertise covers the entire development: (i) sequencing of antibodies, (ii) design of proteins, (iii) constructions of expression vectors, (iv) production in eukaryotic expression system (CHO, without serum ), (v) automated purification and (vi) structural and functional characterizations (using recent techniques such as nanoDSF or thermophoresis). Many optimizations can also be performed on "antibodies" in order to improve their developability. Expertise in custom design and development of recombinant proteins.

Equipment: all the necessary equipment for culture (incubators, shakers, microbiological safety station, etc.), molecular biology (thermal cyclers, electrophoresis tanks, ultracentrifuge, etc.), biochemistry (tanks for SDS-PAGE, devices for tip for exclusion-diffusion chromatography, aggregation, interaction, stability, affinity measurements: ÄKTA Pure, Monolith, Prometheus).

INRAE DOVE research Team:

The "Egg Defense, Development, Evolution" (DOVE) team is one of the 4 teams of the "Bird Biology and Aviculture" (BOA) of the INRAE ​​Val de Loire Center (Nouzilly). The team brings together skills on the different structures involved in egg defense and its quality (species of agronomic interest: hen / chicken, duck and guinea fowl, in particular). Its scientific projects cover both fundamental research and applied research for the "eggs for consumption" and "eggs for hatching" sectors. The DOVE team also benefits from the expertise of the 3 other UMR BOA teams in Physiology, Nutrition, Genetics, Biostatistics and Metabolism of farm birds, from projects with the Technical Institute of Aviculture (ITAVI) within the framework of of the Joint Technological Unit BIRD, and the geographical proximity of the Tours Poultry Experimentation Center (https://doi.org/10.15454/1.5572326250887292E12).

Expertise in "omics" data analysis (transcriptomics, proteomics), in the physiology of egg formation, on the mechanisms involved in biomineralization (shell), its quality criteria and on the role of the egg during Embryonic development. The team relies on collaborations with the CIRM-BP and 4 to 5 teams from the UMR Infectiology and Public Health for questions relating to immune defense and host / pathogen interactions, the Center for Molecular Biophysics (CNRS, Orléans) for structural analyzes of antimicrobial proteins, Proteomics platforms (PIXANIM, INRAE, Tours), electron microscopy (IBISA Electron Microscopy Platform, University of Tours) and metabolomics (Metabolomics and Chemical Analysis Platform, University de Tours) for integrated molecular, physicochemical and macroscopic approaches. The team maintains long-standing collaborations with national and international teams (Canada, Spain, Germany, Great Britain, in particular) dedicated to understanding the mechanisms associated with bird egg formation and genetic variability. , structural, biochemical and molecular of the different defense systems of the egg. The team also conducts research in collaboration with private sector companies in the fields of animal nutrition and genetic selection. The members of the team are involved in 3 learned societies (World Poultry Science Association, French Society of Biomineralized Tissues, GDR MultiFunction of Antimicrobial Peptides).

Equipment : DET6000 and Instron for egg quality measurements (shell, internal compartments) / equipment for protein and peptide purification (affinity and reverse phase chromatography columns, electrophoresis) and for Q expression study -RT-PCR (382 wells).

EBI - Laboratory of Ecology and Biology of Interactions (EBI) - UMR CNRS 7267

EBI is developing research on the interactions between host species, microorganisms and environmental factors. These themes are addressed through an integrated approach that goes from the molecule to the organism and to ecosystems. Environmental biology, environmental quality, water microbiology, environmental genomics.

Equipment : Biotron Plateau: experimental devices for controlled environments for invertebrates and plants; Enviromics platform: devices intended for “omics” and bioinformatics techniques; Microbios platform: devices for analyzing microorganisms in the environment.
